"Most assuredly" - this is the language of one who has true knowledge of all things: God Himself in the Person of Christ.

Although Nicodemus was "the teacher of Israel", he had difficulty in following the Lord's statements. His thoughts were limited to what someone can perceive with one's natural senses. Nicodemus could not conceive of regeneration as it had been indicated in the Old Testament.
